G protein-coupled receptor activated by the neuropeptide pituitary adenylate cyclase-activating polypeptide (ADCYAP1/PACAP).
This gene encodes type I adenylate cyclase activating polypeptide receptor, which is a membrane-associated protein and shares significant homology with members of the glucagon/secretin receptor family. This receptor mediates diverse biological actions of adenylate cyclase activating polypeptide 1 and is positively coupled to adenylate cyclase. Multiple alternatively spliced transcript variants encoding distinct isoforms have been identified.

Function. G protein-coupled receptor activated by the neuropeptide pituitary adenylate cyclase-activating polypeptide (ADCYAP1/PACAP). Binds both PACAP27 and PACAP38 bioactive peptides. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors. Activates cAMP-dependent pathway. May regulate the release of adrenocorticotropin, luteinizing hormone, growth hormone, prolactin, epinephrine, and catecholamine. May play a role in spermatogenesis and sperm motility. Causes smooth muscle relaxation and secretion in the gastrointestinal tract.
Subunit / interactions. Interacts with maxadilan, a vasodilator peptide from Lutzomyia longipalpis saliva; the interaction results in ADCYAP1R1 activation.
Subcellular location. Cell membrane.
Tissue specificity. Most abundant in the brain, low expression in the lung, liver, thymus, spleen, pancreas and placenta.
Activity regulation. Several synthetic peptides derived from maxadilan, a vasodilator peptide from Lutzomyia longipalpis saliva, act as antagonists for ADCYAP1R1.
