WFDC2 (WAP four-disulfide core domain 2, HGNC:15939) is a protein-coding gene on chromosome 20q13.12, encoding WAP four-disulfide core domain protein 2 (Q14508). Broad range protease inhibitor.

This gene encodes a protein that is a member of the WFDC domain family. The WFDC domain, or WAP Signature motif, contains eight cysteines forming four disulfide bonds at the core of the protein, and functions as a protease inhibitor in many family members. This gene is expressed in pulmonary epithelial cells, and was also found to be expressed in some ovarian cancers. The encoded protein is a small secretory protein, which may be involved in sperm maturation.

Disease relevance. Bronchiectasis and nasal polyposis (BENP) [MIM:620984] An autosomal recessive destructive airway disease characterized by persistent abnormal dilatation of the bronchi, and severe chronic rhinosinusitis with nasal polyposis. Affected individuals suffer from sub-normal lung function, chronic productive cough, excessive sputum production, and recurrent sinopulmonary infections. The disease is caused by variants affecting the gene represented in this entry.
