Summary

WWC3 (WWC family member 3, HGNC:29237) is a protein-coding gene on chromosome Xp22.2, encoding Protein WWC3 (Q9ULE0). Regulator of the Hippo signaling pathway, also known as the Salvador-Warts-Hippo (SWH) pathway.

This gene encodes a member of the WWC family of proteins, which also includes the WWC1 (KIBRA) gene product and the WWC2 gene product. The protein encoded by this gene includes a C2 domain, which is known to mediate homodimerization in the related WWC1 gene product.

Function. Regulator of the Hippo signaling pathway, also known as the Salvador-Warts-Hippo (SWH) pathway. Enhances phosphorylation of LATS1 and YAP1 and negatively regulates cell proliferation and organ growth due to a suppression of the transcriptional activity of YAP1, the major effector of the Hippo pathway.

Subunit / interactions. Forms homodimers and heterodimers with WWC1 and WWC2. Interacts with DLC1 and PRKCZ. Interacts (via WW domains) with LATS1 and LATS2.

Subcellular location. Cytoplasm. Cytosol.

Similarity. Belongs to the WWC family.
